Dr Soomro was offering his morning prayers in a madrassa in Gulshan Iqbal Society in Sukkur when unidentified gunmen entered the mosque and shot him dead.
"Two armed men entered in the mosque and fired 11 shots, four bullets hit Khalid Soomro and he died on spot," Tanveer Hussain, a senior police officer told AFP.
Police have since prepared a report which revealed that the assailants who arrived in three cars and entered from the back gate.
It is not yet known whether he had been receiving threats or if he had any enmity with any persons, however, witnesses at the scene have been questioned and police have recorded their statements.
No group has claimed responsibility for the attack but JUI-F has previously been targeted by the Pakistani Taliban, even though the party leaders have acted as negotiators between the militants and the government on several occasions.
Soomro was immediately taken to the hospital but reportedly passed away before his arrival. No other persons were attacked or harmed.
His body was taken to Larkana for burial this evening.
Khalid Soomro was an influential member of the party, especially in the Sukkur and Larkana areas. He had served as a member of Pakistan's senate (upper house) from 2006 to 2012.
Prime Minister Nawaz Sharif took notice of the attack and expressed his sorrows. A strike will be held in various parts of Sindh to mourn the death.
Several areas have also been put on high alert and security checks are being carried out.
JUI-F leaders have often been subjected to brutal attacks, especially in the past few years.
Much recently, JUI-F Chief Maulana Fazlur Rehman narrowly escaped a suicide attack in which he was targeted. He managed to remain unhurt.
He also survived a suicide attack in October after addressing a rally in Quetta in which at least 11 people were killed and 30 wounded.
Responding to the attack, JUI-F Chief Maulana Fazlur Rehman in a press conference in Islamabad announced a nationwide protest on November 30.
"A protest is our right. If people can hold rallies then protesting for a murder is our right and we will have it tomorrow," said the chief.
He condemned the attack and said that the killers should be handed over to them as the state does not do enough for the protection of its people.
"Tell me is there any law in Pakistan?" he questioned.
The chief will be travelling to Sukkur tomorrow.

He was moderate religious leader, always spread the message of tolerance and peace. His assassination should be an eye opener for federal and provincial govt that Sindh is also no more safe from extremists. Terrorists are extending their reach to Sindh as well. A moderate, federalist and pro-people political figure from Larkana Sindh is no more with us, a big loss for Pakistan.
Is it a coincidence that only the opponents of PTI and PMLN are targeted?
Now why would any one take the task of murdering a religious man who ay have been from a different insitution? It took the European Nations 30 years to fight it out among the different branches of christianity which came to end when the west phalien agreement was signed, allowing each Nation whether small or large to practice their christian fath as they see it fit. In muslim lands it has recently appeared on the agenda and thirty years is a pretty long time.

@Malik: there are strong reason behind this killing-nurseries are no more required-these elective religious parties have only divided us by spreading other desired against Pakistan"s people interest and their religious belief.
Strong massage was given earlier too that there is no more space for any more spreading of other thoughts in Pakistan.We already had enough of religious parties influence in the policy making institutes during the last four decade which could not unite us therefore so now this killing should be taken seriously. Inter-faith and inter-sect harmony is order of the day.
Keen to hear what Maulana Fazlur Rehman has to say.. I'm sure he won't have the guts to unequivocally condemn the actual perpetrators, but blame American, Jewish & Indian agents, just like Qazi Hussain Ahmed...
While a lot of people do not have much sympathy for JUI-F, especially PTI people of late, we should remind ourselves that a loss of human life is not a joke.
A murder is a murder!
When you choose to remain silent when others are killed in the name of religion. When you choose to remain silent when minorities are persecuted under the blasphemy law. When you choose to remain silent when women are badly discriminated against. Your choice is wrong. I say..... this killing is wrong and the government has miserably failed in its duty.
Clearly TTP is still active and can carry out such activities at will. Army, together with police, administration, Government and whole nation has to work in unison to clean the menace and unarm the country.
