The oil tankers were en route to Afghanistan from Karachi when four unidentified men riding on two motorcycles stopped them and opened fire.
The drivers fled the scene as soon as the assault began.
The fire brigade, police and rangers have cordoned off the area and the fire has been extinguished.
Earlier last month, a group of 30 armed men attacked Nato oil supply tankers at the national highway near Khairpur, wounding four people and destroying ten vehicles.
According to reports, the oil tankers, carrying fuel for the Nato forces in Afghanistan, were parked at a highway hotel. A group of more than 30 armed men opened indiscriminate fire at the tankers, causing them to catch fire. The assailants disappeared in the darkness.
Ten oil tankers were gutted, while the district’s fire brigades managed to save eight from catching fire.
