The polling station where the incident occurred is located in the Orangi Town area where by-elections for PS-95 are scheduled to be held on August 22.
The two injured policemen from Maripur police station, who had been deployed at the station for election duty were identified as Abu Sufiyan and Abdul Khalil.
Following the incident, extra contingent of law enforcers reached the site and shifted the injured to the Abbasi Shaheed Hospital for medical treatment.
Police officials while quoting the initial investigation said that at least two armed men were behind the incident.
No case was registered till the filing of the report.
Earlier, the Election Commission of Pakistan (ECP) said that it had finalised a security plan for the by-elections which will be in effect from August 21-23.
A large number of Pakistan Army soldiers have already been deployed in three constituencies of Karachi ahead of by-polls to ensure security.
The provincial government has also imposed a ban on carrying and displaying weapons for the duration of by-elections.
The decision was taken at a meeting of the electoral body chaired by the acting chief election commissioner, Justice Tassadduq Hussain Jilani on Wednesday.
