After being put on bat first, Zalmi managed to post 157 in their allotted 20 overs, courtesy Liam Dawson's stunning 62 off 35.
For Gladiators, pacer Rahat Ali was the best bowler on show as he claimed four wickets.
Sublime Ronchi inspires Islamabad to PSL3 final
In reply, Gladiators were on course for an easy win, but cluster of wickets in the middle left the team with 25 to get in the final over.
All-rounder Anwar Ali smashed three maximums to leave the team with three to get off the final ball.
On to the last delivery of the match, Anwar was dropped at the mid on boundary by Umaid Asif as he attempted to win the match for his team with a six, and then his non-strike partner Mir Hamza was run out in an attempt to complete the second run, which gave Zalmi a one-run victory on the night.
